Screen, Select, Enhance: A joint evitria AG – Biofidus workflow for ADCC‑enhanced antibody development

We collaborated with evitria AG on an integrated workflow that connects evitria’s high‑throughput antibody expression and selection pipeline with mechanistic validation for ADCC‑enhanced candidates.

The poster, authored by Julia Pizzolato (evitria AG) with contributions from our colleagues Julian Wichmann and Tobias Luttermann, reports that evitria’s in-licensed GlymaxX‑mediated afucosylated antibody technology selectively increase FcγRIIIa binding (including a 4.4‑fold decrease in K_D), while maintaining target engagement and showing no impact on FcγRI binding.

In contrast, the LALAPG variant strongly reduces FcγRIIIa binding (>300‑fold) and diminishes FcγRI binding to non‑detectable levels; importantly, combining GlymaxX and LALAPG enables controlled tuning of effector function across a wide dynamic range.

Comparable results in a mouse IgG2a background (including a reported 1800% affinity increase with GlymaxX) support cross‑species applicability and help de‑risk translational studies.

The data demonstrate how the evitria–Biofidus collaboration closes the loop from candidate generation to functional, ADCC‑relevant evidence within a coordinated workflow—linking evitria’s HTP‑to‑lead development with Biofidus’ mechanistic characterization.
